Item In Vitro Investigation of the Anticancer Activity of Friedelin in Glioblastoma Multiforme(Afyon Kocatepe University Journal of Science and Engineering, 2018-11-28) Emsen, Bugrahan; ASLAN ENGİN, TUBANUR; Turkez, HasanThere are different herbal methods used for support in many cancer diseases. Lichens are important organisms containing unique herbal compounds and it is known that they have different anticancer activities. Starting from these features, the present study was aimed to investigate anticancer activity of friedelin (FRI), a lichen compound against glioblastoma multiforme (GBM) showed dangerous malignant properties within brain cancer species. It was used human U87MG-GBM cancer cell lines and primary rat cerebral cortex (PRCC) non-cancerous cells isolated from Sprague-Dawley rats in order to determine side effect level of FRI. In the experiments, cytotoxic (via 3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide (MTT) and lactate dehydrogenase (LDH) tests), antioxidant (via total antioxidant capacity (TAC) test), pro-oxidant (via total oxidative stress (TOS) test) and genotoxic (via 8- hydroxy-2′-deoxyguanosine (8-OH-dG) test) activities of different concentrations of FRI were tested. As a result of the study, MTT assay revealed that FRI showed higher cytotoxic activity on U87MG cells compared to PRCC cells (median inhibitory concentration (IC50): 46.38 and 1271.77 mg/L, respectively). Based on U87MG cells, it was determined a significant positive correlation between LDH and TOS activities. High positive correlation between TAC and cell viability on healthy PRCC cells exhibited antioxidant capacity of FRI. Consequently, the results obtained from the present study proved the potential of natural product with an antioxidant capacity as a source for anticancer compound against GBM.Item Inhibition of growth of U87MG human glioblastoma cells by Usnea longissima Ach.(Anais da Academia Brasileira de Ciências, 2019-10-07) EMSEN, BUGRAHAN; OZDEMIR, OZLEM; ASLAN ENGİN, TUBANUR; TOGAR, BASAK; CAVUSOGLU, SEYDA; TURKEZ, HASANHerbal medicines are efficient to reduce side effects in the fight against glioblastoma, which plays a critical role within brain cancer species. The recent studies designated for testing the effects of lichens that have shown numerous anticancer activities on glioblastoma so far. In the present study, different concentrations of water extract obtained from Usnea longissima Ach. were used in order to determine cytotoxic (via 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and lactate dehydrogenase tests), antioxidant (via total antioxidant capacity test), pro-oxidant (via total oxidant status test) and genotoxic (via 8-hydroxy-2′-deoxyguanosine test) effects of them on human U87MG-glioblastoma cancer cell lines. Primary mixed glial-neuronal non-cancerous cells from Sprague-Dawley rats were also utilized to measure the effects of treatments on non-cancerous cells. Based on median inhibitory concentration values, the data belonged to non-cancerous cells (2486.71 mg/L) showed distinct towering compared to U87MG (80.93 mg/L) cells. The viability of non-cancerous and U87MG cells exposed to extract is decreased in a dose dependent manner. It was also showed that low concentrations of extract notably increased total antioxidant capacity on non-cancerous cells. In addition, various phenolic compounds in extract were detected through high-performance liquid chromatography. The recent results encourage that extract will be able to have therapeutic potential against glioblastoma.Item Standing of biotechnology subjects found in biology courses of highereducation and profiling of prospective teachers for their interests on biotechnology(Frontiers in Life Sciences and Related Technologies, 2021-04-30) ASLAN, ALİ; ASLAN ENGİN, TUBANUR; KURMANBEKOVA, GÜLBÜBÜ; KAYALAR, FETHİ; KAYALAR, FİLİZ; KARAGÖZ, YALÇINIn this study, profiling of prospective teachers for their interests on biotechnology in higher education institutions has been conducted and in conjunction with this, the extent of biotechnology subjects in the textbooks of higher education institutions has been investigated. A questionnaire has been prepared to reveal the educational level on biotechnology in higher education institutions and the interest whom prospective teachers have in biotechnology. The prepared questionnaire has been applied to 100 prospective teachers receiving education in the Departments of Physics, Chemistry, Biology, and Applied Sciences Education in Kazim Karabekir Faculty of Education at Ataturk University. As a result of the questionnaire applied to prospective teachers, it has been determined that 54 of the prospective teachers are fond of the subjects related to biotechnology. Also, the assessment carried out for showed that most of the prospective teachers (65) recognize the importance of biotechnology but also they believe that the subjects of biotechnology in higher education are not covered enough and the information given is not sufficient and the necessary explanations are not made about its importance.Item Usnea Longissima Ach. Liken Türününün Metanol Ekstresinin İn Vitro Sıçan Midesinde Mide Asit Sekresyonu Üzerine Etkisi(Atatürk Üniversitesi Vet. Bil. Derg., 2017-12-20) ASLAN ENGİN, TUBANUR; ÇELEBİ, FİKRET; ŞENGÜL, EMİN; GELEN, VOLKAN; ÇINAR, DURSUNALİLikenler alternatif tıpta yaygın olarak kullanılanılan birliklerdir. Likenlerin biyolojik içerikleri ve etkinlikleri üzerine çok sayıda araştırma bulunmaktadır. Bu araştırmada Usnea longissima Ach. liken türünden elde edilen metanol ekstresinin in vitro sıçan midesinde asit salgısı üzerine etkisinin araştırılması amaçlanmıştır. Çalışmamızda Sprague-Dawley ırkı 24 adet erişkin erkek sıçan kullanıldı. Sıçanlar anestezi altında ötenazi edildiler. Mide dokusu izole edildi ve serozal solüsyon bulunan petri kabı içinde temizliği yapıldı. Mide içeriğinin boşaltılması işlemi ise mukozal solüsyon ile yapıldı. Usnea longissima Ach. metanol ekstresinin mide asit salgısı üzerine beş farklı dozunun (0.012 µg/ml, 0.12 µg/ml, 1.2 µg/ml, 12 µg/ml, 120 µg/ml) etkileri, izole organ banyosunda nöronal (asetilkolin) ve parakrin (histamin) yolakların antagonistlerinin varlığında ve yokluğunda araştırıldı. Sonuç olarak, Usnea longissima Ach. ektraktının farklı dozlarının mide asit sekresyonunu önemli düzeyde artırdığı belirlendi. Usnea longissima Ach.’nın mide asit salgısını artırıcı etkisinin histamin aracılı parakrin yolak üzerinden olduğu kanaatine varıldı.
